I love new methods that make everything I learned in grad school obsolete. NEB HiFi cloning 3-fragment assembly scaled down to 6-ul reactions transformed into 1/4 of a โ€œsingleโ€ comp cell aliquot in strip tube. 300 ul SOC, streak 100 ul on a plate, and pick two colonies. Send one for whole-plasmid sequencing (Primodium or Plasmidsaurus) without screening. Perfect.
