CryoSPARC now has several fancy algorithms to sort heterogeneity. But the more #cryoEM datasets I process, the more I believe that 3D classification (with no alignment) and local refinement are all we need. Combined with some understanding of the particle under study and carefully crafted masks, they are applicable to (and in fact solve) so many cases.

Today's Friday project is revisiting a dataset that has been sitting around since the first pass analysis had mostly convinced us that it didn't contain the complex we were after. Keyword "mostly". Revisiting this dataset now by 3D classification under new assumptions (occupancy of the component of the complex we are looking for may well be much lower than expected from the biochemistry). Don't want to jinx it until I see a refined map, but it starts looking like I found that little protein! 🤩

@naturepoker @thatdnaguy ha ha don't worry, data storage is also a hot mess in "professional" labs! 😅

The average academic group doing #cryoEM experiences exactly what you describe. The bottleneck is either storage, or how fast we can complete projects to the point when we can deposit datasets to a public database and free up storage space on our end.

The storage space used now to hold data for projects in the sloooow late phases of writing up and submitting, is literally preventing me from starting new projects... (which, in a way, is good when trying to finish projects... negative feedback loop and all that).
