The movie shows 2 cell populations, stained separately with our SPY-DNA probes, that were subsequently mixed together and further stained with SPY700-FastAct, allowing to track the 2 populations based on the color of the nuclei.

All of this started because I wanted an excuse to do what we lovingly refer to as the puffs assay - high-speed TIRF imaging of pH-sensitive GFP-tagged receptors that allows us to visualize receptor recycling as bright puffs of fluorescence.
