Mutualistic interactions can drive processes of diversification. To understand their evolution, one has to compare symbionts with free-living relatives. Knowing which these are and how they adapted to their environment may hint at what has triggered a symbiotic shift. Fungus-growing ants (subtribe Attina) mostly cultivate two clades of fungi from the genera Leucoagaricus and Leucocoprinus, but as in many cases, the free-living fungal ancestors remain unknown. Here, we reconstructed comprehensive phylogenies (ITS and/or LSU) for 179 new ant-fungal samples, 310 free-living fungarium samples, and 323 ant-fungal samples and 493 free-living samples from GenBank, totalling 1,305 specimens. We then selected 354 specimens for phylogenetic tree reconstruction at species level and investigated trait evolution and branch clustering patterns using MBASR and RPANDA. We show that the classic division of ant-fungi in two clades is an underestimation and that leucocoprinaceous fungal farming arose at least four times. Furthermore, reversals from mutualism to free-living fungi happened at least once. Finally, the genera Leucocoprinus and Leucoagaricus are recovered paraphyletic, sometimes with other genera such as Lepiota spp. included in the clades. These results highlight the current taxonomic generic delimitations within the Leucocoprineae are in need of a thorough revision since they are hindering the understanding of the ecological and phenotypical evolution of the fungus-ant mutualism.
