Emergent biosynthetic pathway to essential amino acids by metabolic metathesis

* alkene metathesis unknown in extant biochemistry
* alkenic amino acid vinylglycine can be generated in biocompatible reaction
* vinylglycine produced in situ then used as precursor of isoleucine & methionine
* allows growth of Escherichia coli strains normally requiring these essential amino acids

#DirectedEvolution is a powerful method but has not been easy to do with gigantic #nonribosomal biocatalysts that make important #antibiotics. We have developed a #microfluidics screen that uses liposomes as sensors for membrane active peptides. Bacteria are grown in tiny droplets. If they make gramicidin S, the sensor liposomes burst, liberate calcein, and generate fluorescence. This was tested for screening mutants and may become a useful tool for directed evolution. biorxiv.org/content/10.1101/20

@davidschnettler and the Hollfelder-team have used #DirectedEvolution to breed new phosphotriesterases that could be useful for bioremediation or treatment of organophosphate intoxication. Active #enzyme variants were retrieved from large libraries packaged in tiny droplets on a #microfluidics platform.

Here, we show that an alternative motif for phosphotriester hydrolysis exists, starting #DirectedEvolution from a metal-independent alpha/beta hydrolase which Pierre-Yves identified using functional #metagenomics:

Phosphotriesters are so toxic because they irreversibly inhibit the catalytic triad of acetylcholinesterase. So, our starting point is a homologue of exactly this target, just with a cysteine instead of a serine in the catalytic triad. (3/6)

In this work carried out during my PhD together with Jamie Klein, Tomasz Kaminski, and Pierre-Yves Colin in the Hollfelder Lab at #UniversityofCambridge we evolve a #phosphotriesterase with a new, metal-independent mechanism by using droplet #microfluidics.

#introduction

Hi, I'm a biochemist enthusiastic about #enzymes, #biocatalysis and #ProteinEngineering & #ProteinEvolution.

I did my PhD with Florian Hollfelder at the #UniversityofCambridge working on #DirectedEvolution of enzymes using droplet #microfluidics. Currently, I am doing a postdoc at #ETHZurich, studying which properties determine a protein’s innovability towards new function.

Looking forward to learning about cool preprints/papers and finding a lively scientific community here!
