Androgen receptor-positive #ProstateCancer cells express hormone-dependent tRNA halves, but what are they for? This study shows that the 5′-tRNALysCUU half promotes #CellCycle progression by destabilizing the mRNA of p21, a negative regulator @PLOSBiology plos.io/3SHIilO

Schematic representation of how the 5′-tRNALysCUU half promotes cell proliferation. Lefthand panel shows the normal situation in AR-dependent prostate cancer cells, with the 5'-tRNA half destabilising p21 mRNA, resulting in lower p21 levels and boosted cell cycle progression. Righthand panel shows cells with reduced levels of 5'-tRNA halves, where the higher levels of p21 result in cell cycle arrect.

What is the role of m5C #RNAmodifications during neural development? This study shows that m5C reader protein Ybx1 plays a crucial role in promoting #CellCycle progression of embryonic cortical #ProgenitorCells, essential for proper cortical development @PLOSBiology plos.io/3SUN4we

Left: Coronal brain sections of control (top), heterozygous (middle) and knockout (bottom) mice at E13.5 were stained with antibodies recognizing BrdU and Ki67. Pregnant mothers received a BrdU pulse 24 h before embryo collection. Representative cortical regions were shown. Scale bar, 50 μm. Right: A model in which the m5C reader protein Ybx1 regulates the stability of transcripts of key cell cycle regulatory proteins, and its loss leads to impaired proliferation and differentiation in cortical progenitor cells. Multiple #myeloma often requires IRE1 to sustain malignant growth, but why? This study shows that IRE1 promotes #CellCycle progression via the transcription factor IRF4, paving the way for new therapeutic strategies @PLOSBiology plos.io/43TnG0L

Hypothetical model for nonenzymatic IRE1 cell cycle regulation via IRF4. Schematic representation of IRE1 supporting IRF4 activity/expression, promoting E2F1 and CDC25A transcription, in turn driving CDK2 activity to enable unconstrained cell-cycle progression. Silencing of IRE1 increases IRF4 phosphorylation on Ser114 and Ser270, which attenuates IRF4’s self-sustaining transcriptional activity, depleting E2F1 and CDC25A and leading to cell-cycle arrest. Phosphoresidues annotated in red: inhibitory. Phosphoresidues annotated in green: activating. Created in BioRender.

ER-resident IRE1 plays a role in #cancer development via enzymatic activation of XPB1 TFs and regulated #RNAdecay. This study shows that some cancer cells also have an unexpected nonenzymatic dependency on IRE1 to promote #CellCycle progression & tumor growth @PLOSBiology plos.io/3EaNFX4

Detection of endogenous Halo-tagged IRE1 and non-tagged Lamin B1 by confocal microscopy. U2OS HaloTag cells, containing a C-terminal HaloTag into the endogenous IRE1 gene locus, were examined. Cells were cultured in the presence of Janelia 646 HaloTag ligand (green), fixed, stained with anti-Lamin-B1 (red) antibodies, and analyzed by confocal microscopy. Left: Merge field of IRE1 (red), Lamin B1 (green), and nucleus (blue) is shown. Scale bar = 5 μm. Right: Close up of the nuclear envelope where each channel is shown separately and merged. Scale bar = 2 μm. Pixel size = 48 nm. Representative image of two independent experiments shown.

From post 5,
Amino acids that are enriched/depleted next to the phospho-site reflect which protein kinase enzymes did the phosphorylation.

Phospho-dawn sees acid-directed sites being phosphorylated (D and E enriched at ZT0),
but by evening it's #CellCycle kinases guided by Proline (P at ZT12-16). 😎

#Shiftwork

Motif plot showing which amino acids are enriched or depleted in the residues next to the phosphorylated Serine or Threonine, which reflects the preferences of the protein kinase enzymes that do the phosphorylation. Those enzymes seem to work in alternate shifts, because acid residues (D/E) flank the rhythmic phospho-dawn sites that are most phosphorylated at ZT0 before dawn. Many of the evening-peaking sites are followed by Proline (P), characteristic of GSK3b and the cell-cycle kinases. (from Figure 5C)
